Determination of protease activity. The test for proteolytic activity involves mixing 1 ml of azocasein solution with 100 ml of the sample that contains the enzyme. The contents of the tubes are mixed and incubated at 35.5°C for 15 minutes. The reaction is stopped by adding 2 ml of 5% TCA. The absorbance of the supernatant is recorded at 345 nm in a UV/Visible spectrophotometer.
